  • Effects of Survivin Antisense RNA on Apoptosis and Reversing Drug Resistance of SGC7901 Cells

    Objective To study the effects of survivin antisense RNA on SGC7901 cell’s apoptosis and chemosensitivity to taxotere, and to investigate its effect on the expression of multi-drug resistance gene-1 (MDR-1). Methods Survivin antisense eukaryotic vector anti-pcDNA3-svv was transfected into SGC7901 cell lines by lipofectamine and positive clones were screened out then. Survivin protein and MDR-1 mRNA were measured by western blot and RT-PCR, respectively. Apoptosis that was induced by anti-pcDNA3-svv was observed by electronic microscope, and the sensitivity of SGC7901 cell to taxotere was examined by MTT. Results The expressions of survivin protein and MDR-1 mRNA in transfected SGC7901 cells both decreased more significantly than that of non-transfected cells (P<0.05, P<0.01), and the indices of MDR of transfection group and non-transfection group were 0.196±0.013 and 3.126±0.019, respectively, at the late phase of apoptosis, which had a significant difference between each other (P<0.01), IC50 of the transfected cells to taxotere was (16.7±1.98) ng/ml and that of the non-transfected cells was (55.7±1.89) ng/ml, which also had a significant difference (P<0.01). Conclusion Surivivin antisense RNA could induce the apoptosis of SGC7901 cancer cell line and could increase the cells’ sensitivity to taxotere, which may help to reverse drug resistance.

  • MDT discussion of a case of solid pseudopapillary tumor of pancreas

    ObjectiveTo improve recognization and effect of surgical treatment for solid pseudopapillary tumor of pancreas (SPTP) through multidisciplinary team (MDT) discussion.MethodsThe clinical data of 1 case of SPTP treated in The Second Affiliated Hospital of Chongqing Medical University were discussed by the MDT. The best operation scheme was selected and the patient was followed-up.ResultsThe preoperative CT scan showed that the occupation on the body and tail of pancreas and suggested it was the tumorous lesions. After the MDT discussion, the preoperative diagnosis was still not completely clear. However, the decision was made to resect occupation. After the operation, the pathological examination showed it was the SPTP. There was no recurrence or metastasis with the follow-up of 6 months.ConclusionsPreoperative diagnosis of SPTP is difficult, especially for atypical SPTP. Through MDT discussion, it can help to reduce misdiagnosis rate and formulate optimal surgical treatment strategy.

  • Feasibility and safety study of daytime laparoscopic cholecystectomy in high-altitude areas

    Objective To explore the feasibility and safety of daytime laparoscopic cholecystectomy in high-altitude areas. Methods Clinical data of patients with gallstones undergoing laparoscopic cholecystectomy admitted to Qinghai Red Cross Hospital between January and December 2023 were retrospectively collected. Patients were divided into daytime surgery group and routine surgery group according to their hospitalization method. The gender, age, ethnicity, American Society of Anesthesiologists classification, comorbidities, postoperative pain, postoperative indwelling drainage tube, postoperative nausea and vomiting, overall satisfaction, length of hospital stay, and hospitalization costs of the two groups of patients were compared. Results A total of 95 patients who underwent daytime surgery and 186 patients who underwent routine surgery were included. There was no statistically significant difference in gender, age, comorbidities, or American Society of Anesthesiologists classification between the two groups of patients (P>0.05). The proportion of Han patients in the daytime surgery group was higher than that in the routine surgery group (69.5% vs. 34.4%, P<0.05). All patients successfully completed the surgery without any serious complication. There was no statistically significant difference in postoperative indwelling drainage tube, postoperative nausea and vomiting, postoperative pain, or overall satisfaction between the two groups of patients (P>0.05). The length of hospital stay [(1.03±0.17) vs. (5.30±1.53) d] and hospitalization expenses [(8104.97±660.65) vs. (8947.82±746.19) yuan] in the daytime surgery group were lower than those in the routine surgery group (P<0.05). Conclusion Daytime laparoscopic cholecystectomy is safe and feasible for patients in high-altitude areas without comorbidities or with stable control of comorbidities, and is beneficial for reducing the medical burden on patients.

  • Meta-analysis of the diagnostic value of LncRNA in tuberculosis

    ObjectiveThe diagnostic efficacy of circulating long non-coding RNA (lncRNA) for tuberculosis was evaluated by systematic review. MethodsData from PubMed, Web of Science, Cochrane Library, Embase, Chinese Medical Journals Full-Text Database(CMJFD), CNKI and WanFang Data were searched. Literatures on the diagnostic value of lncRNA in tuberculosis from the database establishment to August 20, 2024 were selected, and the quality of literatures was assessed using QUADAS-2 tool. Meta-Disc 1.0 software tested the threshold heterogeneity of the included studies. Stata18.0 software calculated sensitivity, specificity, positive likelihood ratio, negative likelihood ratio, diagnostic odds ratio and other effect sizes, and performed subgroup analysis and meta regression to explore the source of heterogeneity. Deeks funnel plot evaluates publication bias. Results A total of 28 case-control studies were included in 14 literatures. The meta-results showed that the combined sensitivity was 0.88 (95%CI:0.81-0.93), the specificity was 0.90 (95%CI:0.84-0.94), and the PLR was 9.05 (95%CI:5.16-15.87). The NLR and DOR were 0.13 (95%CI:0.08-0.22) and 67.96 (95%CI:27.27-169.39), and the AUC were 0.95 (95%CI: 0.93-0.97). Subgroup analysis showed that lncRNA was more effective in the diagnosis of tuberculosis when PMBC samples, LncRNA expression was down-regulation, the study sample size was ≤100, there was cut-off value, GAPDH was used as the internal reference, and RNA extraction kit was used. meta regression indicated that lncRNA expression level and sample size were the main sources of heterogeneity. Conclusions lncRNA has high accuracy in the diagnosis of tuberculosis, and is expected to become a new biomarker to assist the diagnosis of tuberculosis.

  • Drafting process of “Clinical practice guideline on traditional Chinese medicine alone or combined with antibiotics for patients with acute upper respiratory infection in children”

    The clinical practice guideline on traditional Chinese medicine alone or combined with antibiotics for patients with acute upper respiratory infection in children was issued by China Association of Chinese Medicine in June 2017. Base on the evidence, consensus and experience, five manage groups, steering committee, consensus expert group, systematic review group, drafting group and secretary group, have promoted the project. The main work process refers to up-to-date clinical practice guideline definitions of institute of medicine and quick recommendations guide of guidelines international network and WHO. In order to provide readers with a better understanding of the guideline, the drafting process of guideline principle, main work process, and basis policy were described in detail in this paper. The guidelines’ scientific characteristics, accuracy and practicability were better exhibited in attempt to enhance its promotion and application.
